It seems like there are updates going out to tablets all the time that break roots. RIM and Amazon have already done it, and now, it looks like our book-loving friends at Barnes and Noble are going to do the same thing, as well.
The bookseller today pushed out an OTA update that breaks both the root and the ability to sideload apps. We've written about the potential B&N's tablets have if they are rooted, and how the tablets are fantastic deals once they are, so this update that kills a lot of functionality will also probably kill a lot of sales to people who want to take advantage of its power.
If you're rooted and don't want to lose your tweaks, you can turn WiFi off to stop the updating process. Obviously that isn't a fantastic solution, but it certainly works for the time it takes developers to find an exploit in this update.
